Coupling growth of E. coli to novel modules of synthetic CO2 fixation cycles
Microbial C1 assimilation is paving the way towards a circular bioeconomy. While multiple synthetic metabolic designs overcoming the limitations of nature were proposed and shown in vitro, their translation to living systems has progressed much slower. Therefore, we designed E. coli strains relying on the bottleneck reactions synthetic CO2 fixation cycles for growth and engineered multiple cycle modules that were not yet part of the in vivo metabolism repertoire. Furthermore, we designed a novel workflow for the machine-learning guided expression level optimisation for pathway catalysts. In the future, the demonstrated modules will serve as basis for microbial CO2 fixation engineering.
