Design and Realisation of the Synthetic Hydroxypropionyl-CoA/Acrylyl-CoA Cycle for Carbon Fixation

Abstract: The goal of this project is to bring a synthetic carbon-fixing cycle, which was designed by metabolic retrosynthesis, to life. The Hydroxypropionyl-CoA/Acrylyl-CoA (HOPAC) Cycle proceeds through a series of acyl-CoA intermediates and consumes two NADPH and three ATP while fixing two carbon, producing the C2 compound glyoxylate, and regenerating its substrate. The project takes an iterative approach to producing a functional cycle. The cycle is run, issues are identified, those issues are addressed, and the cycle is run again to determine what, if any, new issues arise.

Functional characterization of type IV CRISPR-Cas systems

Type IV CRISPR-Cas systems are known to be the “mysterious” CRISPR systems, these systems are often found missing adaptation modules and nucleases conduct target cleavage. In this project we identified a type IV CRISPR-Cas system shows PAM-dependent interference activity in vivo. The interference inhibit antibiotic resistance carried in the targeted plasmids. This interference depends on the putative helicase activity of DinG.
