The first step of anaerobic degradation of aromatic hydrocarbons toluene and 2-methylnaphthalene are catalyzed by the glycyl radical enzymes benzylsuccinate synthase (BSS) and newly discovered naphthyl-2-methylsuccinate synthase (NMS) which catalyze the radical addition of toluene and 2-methylnaphthalene to fumarate, producing benzylsuccinate and naphthyl-2-methylsuccinate. We developed the first recombinant overproduction system of active benzylsuccinate synthase and naphthyl-2-methylsuccinate synthase. Also, we present initial data on the effects of some constructed active-site mutants in the alpha subunit of BSS and initially characterize substrate spectrum of NMS. This study provides new insight on the reaction mechanism and substrate recognition of both enzymes.
