About the creation and isolation of switching metabolic enzymes

Bioprocess engineering often involves the deregulation of overproduction pathways, leading to higher production rates but at the same time draining resources from native metabolism. This can result in cells that are less robust against stress. Synthetic allosteric regulation of enzymes of overproduction pathways could solve this problem by allowing the cells to regulate the fluxes internally dependent on the growth conditions. In my project, the goal is to create such enzymes with synthetic allosteric regulation. As the used methods result in large libraries of possibly switching enzymes, we furthermore developed a FACS-based screening method to identify and isolate switching enzymes.
